Will exclusion get rid of pests I already have?

Exclusion prevents entry, so it is most powerful paired with treatment when an infestation is active — we remove the current population, then seal so it cannot recur. On a pest-free home it is purely preventive. Either way, sealing the building is what makes the result permanent instead of a temporary knockdown.

Is pest-proofing a one-time job or ongoing?

It is largely a one-time investment that keeps paying off, though buildings settle and develop new gaps over time, so periodic re-inspection is wise — especially after renovations or weather damage.
